Please help me in understanding How the assignment knows that it is completed ?

To add a bit more technical detail, the assignment itself is a separate object in the database and when the work item advances off of the assignment shape in the flow, the old assignment is deleted from the database. A new assignment object is added to the database if the work object lands on a new assignment shape.

Whenever the flow action which is connected to the assignment shape in flow is successfully exicuted/submitted then Wor Item not be in the Current Assignment
